Roads in the city centre, Edgbaston, Stirchley, Bournville and Selly Oak will be completely or partially shut from late Saturday night until Sunday afternoon.
More than 12,300 runners are taking part in the EDF Energy Birmingham Half Marathon and thousands more will line the streets to support them.
Also being held on the same day are the IAAF World Half Marathon Championships which will see around 200 athletes taking part.
The course starts on Cambridge Street, near Paradise Forum, and finishes in nearby Broad Street.
The 13.1 mile-route will go through the markets area before heading down on to Pershore Road, doing a lap of Cannon Hill Park and then rejoining Pershore Road.
From there it continues along residential roads through Stirchley, Bournville and Selly Oak before rejoining Pershore Road, heading back towards the city.
In the city centre the race will continue into Hinckley Street, Hill Street, Navigation Street, Holliday Street and Granville Street before turning into Broad Street where it will finish nearCentenary Square. The World Half Marathon Championships will start at 9am, with the main race at 10am.
All the roads affected will close at 7am on Sunday and not re-open until 4pm. Those on the main route will be shut completely while surrounding streets will only be partially closed.
In addition, Broad Street will be closed to buses from Saturday at 7pm for preparations and this will also impact on services that go to the NIA.
People living in residential streets affected by the closures will have been notified already, but everybody else is asked to check the website www.birminghamhalfmarathon.com to see a full list of the closures.
